Pesticide Intermediate N-Methylformamide With Short Name NMF
N-Methylformamide (NMF) is a colorless, nearly odorless, organic compound and secondary amide with molecular formula CH3NHCHO, which is a liquid at room temperature. NMF is mainly used as a reagent in various organic syntheses with limited applications as a highly polar solvent.
NMF is closely related to other formamides, notably formamide and dimethylformamide (DMF). However, industrial use and production of NMF are far less than for either of these other formamides. DMF is favored over NMF as a solvent due to its greater stability. Annual production of NMF can be assumed to be significantly less than the production of either formamide (100,000 tons) or DMF (500,000 tons).

N-Methylformamide is a water-soluble organic solvent. As an adjuvant antineoplastic agent, N-methylformamide depletes cellular glutathione, a key molecule involved in the antioxidation of reactive oxygen species (ROS) and other free radicals, thereby enhancing ionizing radiation-induced DNA cross-linking in and terminal differentiation of tumor cells. Preparative Methods: N-methylformamide may be produced by mixing formic acid,which forms methylammonium formate. This intermediate is dehydrated using molybdenum trioxide in aqueous. Volatile side products are distilled away at temperatures up to 190 °C to give N-methylformamide in high purity (>99.3%) and in high yield (>92%) (eq).
N-Methylformamide may also be formed by amination of methyl formate.
Purification: may be purified by distillation. May also be dried over freshly ignited 0.4 nm molecular sieves and filtered before use.
Handling, Storage, and Precautions: N-methylformamide is not listed as a hazardous material. It is stable at room temperature. It may be slightly teratogenic and can cause liver damage if ingested or inhaled. N-Methylformamide should be stored in a tightly sealed container in a cool place. Medical studies have shown N-methylformamide to have antitumor activity. This reagent has also been shown to have severe hepatoxic effects, however, due to a causal depletion of hepatic GSH. Normal care should be taken when working with this reagent.
